No. Buy Now buttons skip Upsell Now's discounted checkout process. If your upsell offers include discounts, shoppers will not be able to receive their discounts if they checkout via a Buy Now button.
For this reason, Upsell Now hides Buy Now buttons by default. If you need to show them, you can do so via the Settings tab in Upsell Now.
